چکیده
The re-isolation of trace anlOunts of virus added to tissue hOInogenates has been taken as a Inodel probleIll in the developIllent of high-resolution Illethods for separating particles. FroIll a review of data on these particles, it was found that, when sediIllentation rates are plotted against their banding or buoyant densities, IllOst viI'uses fall in a "window" which appears to be free of other particles. To exaIlline this concept experiIllentally, a technique for sequential rate-zonal and isopycnic-zonal centrifugation has been developed which allows this two-diIllensional separation to be Illade and visualized. By use of this so-called Sop SysteIll, trace quantities of T3 phage particles have been recovered and identified froIn rat liver, brain, and spleen hOlllogenates. The Inethod Inay be used to isolate trace quantities of virus-like particles that have occasionally been reported in hUInan tunlOrs.-Nat Cancer Inst Monogr 21: 253-283, 1966.

Isolation of native glycogen by combined rate-zonal and isopycnic centrifugation.
Rat liver glycogen was separated into fractions of uniform particle size by the rate-zonal centrifugation of homogenates through cesium chloride and sucrose density gradients. Glycogen was then isolated from other cell constituents having similar sedimentation coefficients by isopycnic-zonal centrifugation in CsCI density gradients to effect a density separation. A density of 1.62 to 1.65 was i...
متن کاملAn introduction to particle separations in zonal centrifuges.
Zonal centrifugation, based either on sediInentation rate or on isopycnic banding in liquid density gradients, is useful for high-resolution separation of subcellular particles. However, the amoun t of material which may be separated is extremely limited when conventional centrifuges are employed. To increase the capacity and, in certain instances, the resolution of the method, the possibility ...
متن کاملAssociation of Lactoferrin with Specific Granules in Rabbit Heterophil Leukocytes
Lactoferrin has been identified in rabbit heterophil leukocytes on the basis of its immunological reactivity, electrophoretic mobility, acid-resistant iron-binding properties, and spectral characteristics. Leukocyte lactoferrin was found to be exclusively localized in the specific (secondary) granules, which have been resolved from other subcellular components by zonal differential centrifugati...
متن کاملSTUDIES ON ISOLATED CELL COMPONENTS XVII. The Distribution of Cytochrome Oxidase Activity in Rat Liver Brei Fractionated in the Zonal Ultracentrifuge
The zonal ultracentrifuge was used to separate the subcellular components of rat liver brei into soluble phase, microsomal, mitochondrial, membranous fragments, and nuclear fractions during a single centrifugafion. The centrifuge was run at 10,000 to 30,000 RPM for 15 to 240 minutes, and the rotor contained a 1200 ml sucrose gradient, varying linearly with radius from 17 to 55 per cent sucrose ...
متن کاملSubcellular distribution of proteins synthesized in slices of rat cerebral cortex.
The incorporation of L-[4,5-3H]leucine into slices of rat cerebral cortex has been measured, with emphasis on the time course of the distribution of label in subcellular fractions prepared by zonal isopycnic centrifugation in continuous sucrose gradients. Rapidly labeled proteins are found in both the soluble and membranous components of the nerve end particles. Except for a minor part of the m...
